How she became Sailor Pluto: No one is really sure how the Outer Senshi (except for Sailor Saturn) got their powers. It is assumed that the Outer Senshi always had their powers. She first appears in Chibi Usa's Luna P ball. She tells the Senshi that they need to rescue Chibi Usa from a dream that she is having. Other than that, I'm not sure how she got her powers. She guards the Gate of Time and isn't allowed to leave her post. She does eventually in Sailor Moon S, Sailor Moon Super S, and Sailor Stars.
Family: As far as I know, Setsuna has no family. There is no mention of them at all. That is one thing about the Outer Senshi, it never says anything about their families.
Friends: Ah, of course her friends are the Senshi. She is more closer with Chibi Usa than anyone else. She is protective of Chibi Usa because Chibi Usa is her only friend who will visit her when she is guarding the Gate of Time. She is also good friends with Hotaru, probably because Michiru and Haruka spend a lot of time together.
Inner Senshi: She has a very good relationship with the Inner Senshi. She helped them out in Sailor Moon R when she first appeared, to help the Senshi find ChibiUsa. She is the oldest, and sometimes likes to work by herself. The Outers in the beginning, don't want to work with the Inner Senshi.
Outer Senshi: She is an Outer Senshi herself and is good friends with all of them. She once gave up her life to save Sailor Neptune and Sailor Uranus in Sailor Moon S, but she returns to life. Setsuna also took care of Hotaru when she was turned into a baby.
Relationships: In the whole series, it was never once said if Setsuna ever loved someone. She is by herself most of the time. I do not think she wants a boyfriend. Her job is to guard the Gate of Time and protect the future queen of Crystal Tokyo. However, there has been some speculation that she is love with King Endymion, though obviously nothing ever came of that.
Past Life: In the Silver Millennium, she was the Princess of Pluto. She watched over Queen Serenity and Princess Serenity from where she was. There is nothing else said.
Future: In the future, she is Sailor Pluto. She guards the Gate of Time and is not allowed to leave her post. She also has to protect Neo-Queen Serenity and her family.
Overall: Setsuna is very wise, mature and reserved. She is extremely knowledgeable, and seems to know a great deal about other time periods. Posing as a Mugen High student, she excelled at science and chemistry as well. She is the oldest of the group, not just due to physical form (human) age, but because she has transcended time in order to guard the gate of time throughout the centuries. It is a solitary job, and she would get lonely. She also struggled with her conscience when the senshi needed her help and she had to choose between helping them and guarding her post. She befriends and watches over Cibi Usa, almost like that of an older sister or mother. She is very loving towards and protective of Chibi Usa. She frequently offers her guidance to Chibi Usa, and to the senshi as well. She even guides Chibi Usa through her Luna Ball when she travels to the past to find Sailor Moon. Setsuna is a bit quiet and mysterious, and rarely shares much about herself. She usually brings messages of what is to come, and helps prepare the senshi to face it. While in Tokyo of the present, Setsuna likes to sit and drink tea with Haruka and Michiru, and has a dream to become a fashion designer, to which her talent for sewing contributes nicely. As Sailor Pluto, she is a dedicated and powerful fighter who has great power. She alone has the power to stop time, but it is taboo, and she is forbidden to use it. She does anyway, in the S season though, to save Neptune, Uranus, and her own life. All in all, Setsuna is a lone protector of the door of time, who watches over the senshi and advises them. She has much wisdom, and shares a special bond with Chbi Usa, the future queen.
