One Stroke painting is painting with a "double-loaded" flat brush. That means you have a light shade and a dark shade of paint on one corner each of your flat brush. After blending the paint into the bristles, you can make strokes that appear to have the highlighting and shading already done, in one stroke, rather than doing these steps the traditional way by basecoating an area in a medium shade, then painting in the darker shading,and then the lighter highlight. Double-loading a brush and painting this way is not new but One Stroke enables you to be able to paint a single petal or leaf in one easy stroke, making the completion of painting a whole flower (or even animals) quicker and easier to accomplish.
Donna Dewberry is the lady that came up with this painting program of her own and was later found at a convention by Plaid. They snatched her up, marketed her techniques, and now she and "One Stroke" are household names to decorative painters worldwide!
