Variations
In addition to regular canasta, there are several special rules that can be added by the players at their leisure.  There are many variations, so not all are listed here. The following is a small list of popular variations. All regular rules apply, with the added twist given.
Canasta History
No Going Out - You cannot meld away or discard your final card. Wait for the stock to be exhausted.

No Freeze - You can't place a wildcard in the discard pile.
Strategic Tips
1 Red 1 Black - Before a team can go out they must have both a red canasta and a black canasta.
Variations of Canasta
Red 3 - You cannot pick up the discard pile until all four red threes have been drawn from the stock.

The following are 4 of my own creations.
No Maddness - If the discard pile exceeds 20 cards in size, you are no longer allowed to pick it up.
World War - You are not allowed to pick up the pile UNTIL it exceeds 20 cards in size.
Underdog - The team in the lead is not allowed to pick up the discard pile until the losing team has made their opening meld.
Tag Team - As in pro wrestling, this variation involves tagging your partner in to the game. Only the member of the team who is 'tagged in' can meld cards. To avoid constant tagging, the players decide before the game how many tags can be made in a single hand.
Other Variations
Dirty Hearts - all hearts act as stop cards. This could be dirty clubs, dirty diamonds, or dirty spades.
No Brainer - you cannot pick the discard pile at all. You cannot go out.
Lucky 7's - Sevens act as additional black threes. (they follow the melding rules of black threes as well)
Red Only - Only red canastas are allowed.
Cracked Mirror - You cannot meld cards of the same rank that your opponents have melded. For example if they meld three sevens, then you meld three sevens, you have cracked the mirror, and lose the game.
