APPLICATION OF THE COMPUTERISED MORPHOMETRIC ANALISYS OF PLATELETS FOR THE QUALITY CONTROL OF PLATELET CONCENTRATES

Monitoring the storage lesion of platelet concentrates (PCs) is commonly done by determining metabolic parameters and functional tests. To date with development of computerized methods of the analysis of the cell's image there is the new opportunity for an estimation of the integrity of platelets during storage. Single donor PCs were prepared by apheresis technology and stored for one or three days at 22 degrees C linearly agitated. Platelets from PCs and from donors whole blood were analyzed morphologically by immersion light microscopy using smears stained by May-Grunwald method in our modification. Area, polarization (P), form factor (FF), share of red and blue colors and the optical density of platelets there measured. Distribution of each parameters were estimated with parametric and nonparamertic statistic criteria. Was received, that after a procedure of preparation of PCs the area and optical density of platelets did not change. Platelets in freshly prepared concentrates were somewhat activated, indicated by pseudopodia formation and had significantly greater polarization and form factor in comparison with whole blood platelets: P=0,236±0,004; FF=15,041±0,206 and P=0,148±0,005; FF=13,363±0,147 correspondingly (Kolmogorov-Smirnov p<0,05). This tendency was kept on of a 3 day of storage. After three days of storage area, share of red color and the optical density of platelets corresponded to normal meanings, although a high proportion of starlike and bizarre platelets was appeared. These results allow to conclude that there is no significant lesions of platelets at the given terms and conditions of storage. So, the computerized morphometric analysis of platelets could serve as an new indicator for the quality of stored PCs.
