Plant leaves inoculated with strains J18 (virulent on Xa21) or J18 (1078) (Avirulent on Xa21) were frozen in liquid N2 after infection at room temperature at 0, 15��, 1, 2, 4, 6, 8, 12, and 24 hours. The RNA was extracted using TRIZ0L reagent (Pharmacia) and quantified by spectrophotometry. Equal amounts of RNA were run on a Formaldahyde Agarose Gel, stained with acryine orange and transferred to a nylon membrane. 32P Radio-labeled DNA from Xb10, WRKY1, WRKY2, and WRKY3 was used as probe to indicate activity of individual WRKY��s. Radioactivity was visualized using a phosphoimager (Molecular Dynamics). |
