Roger Stoneburner as The Joker

Assuming they're following the plot of the first Batman film, the Joker was once Jack Napier, a stooge for a prominent Gotham City crimeboss. During a "job" at a chemical factory, Batman arrived and Napier fell into a vat containing a strong chemical. When he crawled his way out, Napier found that his skin had been bleached white, and his face frozen in a smile. This drove him mad, and he became the Joker, the arch-nemesis of the caped crusader.
As the war between Batman and the Joker's criminal factions raged on, it eventually reached a boiling point. Batman and his partner Batgirl brought down the Joker's operations, resulting in a showdown between the two sides. Batman let the Joker live, and he retaliated by shooting Batgirl, and murdering Catwoman, with whom Batman had a daughter. Batman left Gotham, and the Joker was later apprehended and sent to prison. The Joker's girlfriend, Harley Quinn, carries on in his stead, trying to rebuilt his criminal empire.

A powerful businessman in New Gotham, Larry Ketterly has the ability to bring about his victims worst fears, driving them mad. Larry used this to cause the deaths of his business partners, in an attempt to regain control of the old Gotham dockyards which had once been the hideout for the Joker. Larry, who was a friend of Selena Kyle, tried to get Helena, who opposed him, to kill herself, but the combined might of the Birds of Prey stopped him, though Larry was rendered brain dead as a result. It was later revealed Larry was working at the behest of Harley Quinn.

as "Slick" Waters
Also known as the "Liquid Guy", Slick was sent by Harley Quinn to eliminate New Gotham's few honest cops by using his water-powers to drown them on dry land. Slick's pursuits sent him after Detective Jesse Reese, who was saved by the Huntress. Later, when Slick went after Reese again, Huntress pulled a flamethrower on him, turning Slick into steam. It's not known if he survived this.

Joe Flanegan as Morton

A detective transferred to New Gotham from the neighbouring city of Blüdhaven, Morton appeared to be an average cop, with an interest in the unknown, and a penchant for headaches. It turned out that Morton's headaches were the result of his own meta-human power that let him become aware of another meta-human's presense, and gave him the ability to use their powers. Morton thought of meta-humans as beneath him, and if he were able to kill them all, he would finally be human. Huntress rallied a squad of meta-humans to confront Morton, which overwelmed him, and he leapt off a rooftop to his death.

as Albert "Al" Hawke
The son of one of Gotham's greatest crimebosses, Al Hawke blames the Black Canary for his father's demise, and has been planning revenge on her for years. When the Black Canary returned to New Gotham, Hawke sent his men after her, and it was only through the timely aid of the Canary own daughter, Dinah, that she escaped Hawke's trap. A fight to the finish broke out between Hawke and the Canary, ending with the Canary's supposed demise. Hawke survived, and while in hospital he was visited by detective Jesse Reese, who it turns out is Hawke's son.
Following a quick recovery, Hawke underwent plastic surgery after several attempts were made on his life. Turning to his son Jesse for protection, Hawke confronted both Huntress and a vengeful Dinah, who ended up risking her own life to save Hawke's. In the end Reese took his father into polic custody, rather than let him return to his old tricks.

Al Hawke's right hand man, Frankie Spitz is also a notorious figure in New Gotham's world of organized crime. When Hawke's arch-enemy the Black Canary returned to New Gotham, Frankie helped capture her and take her to Hawke. Frankie was eventually brought to justice, though the Canary died in battle. Sometime after Hawke underwent plastic surgery, Spitz put out a hit on his boss's life. When Frankie tried to call the hit off, the assassin, a man called the Specialist, murdered Frankie and went ahead with it anyway.

Shane Mikael Johnson as Fox of the Animal Gang

A gang of daredevil thrillseekers, who use animal names and masks as their MO for committing robberies. Huntress infiltrated their gang to try to bring them down and was codenamed "Cat". However, Fox, the leader of the gang, lied to Huntress about the gang being non-violent, resulting in the critical injury of Det. MacNally, Jesse Reese's partner on the NGPD. With the aid of Reese and Dinah, Huntress brought down the gang, who were secretly working for Harley Quinn.

Sung Hi Lee as Lady Shiva

as Lady Shiva
Years ago the deadly assassin Lady Shiva appeared to parish in battle with Batgirl. She survived and has returned to get revenge on her arch-foe, who she blames for the death of her sister. Unaware that she is now paralyzed, Shiva framed Batgirl for several murders, while maintaining the secret identity of Sandra Woosan, an old friend of Huntress'. Eventually the truth was revealed and Shiva was presumably taken into police custody.
