Genome analysis of CANOLA (Canada Oil Low Acid), rapeseed oil low in glucosinolate and low in erucic acid ( double 00 Quality)
Activities in the project are:
I. Molecular genetics and genome analysis: DNA extraction, PCR technique ( AFLP, RAPD, SSR markers), developing intevarietal substitution lines, near isogenic lines (NILs), using backcrossing program and marker assisted selection (MAS), genetic map, QTL mapping for quality traits, QTL mapping for agronomically important traits such as flowering time ( begin, end and duration of flowering), winter survival, plant height, disease resistance.
Technics used to detect DNA fragments were an automated DNA sequencer (LI-COR 4200 IR2, LI-COR Inc. Nebraska, USA: real-time laser fluorescence at 800 nm) and also some experience on ABI 3100 Automated Capillary DNA Sequencer II. Tissue culture: microspore tissue culture and double haploid production, plant biotechnology lab.
III. Quality analysis: using NIRS and GC methods to determine fatty acids, oil, protein, glucosinolate, total sinapine (sinapoylglucose and other sinapine esters) and total phytosterols ( campestrol, sitosterol, brassicasterol and etc) contents in the seeds
IV. Field experment:Data collection for agronomically important traits in the field trials, statistical analysis of wide range of phenotypic data from five different locations and years
