1. HPLC gradient shall be run at 214 nm detection on one of the following: (a) C-18 reversed phase column, 3 micron particle size, 4.6 mm width X 150 mm length, 100 Angstrom pore size, flow=1.0 ml/minute, gradient=95%A/5%B->0%A/100%B in 30 minutes (Buffer "A"=99.9% Type I Deionized Water and 0.1% TFA (Trifluoroacetic Acid) and Buffer "B"=99.9% HPLC or nanograde Acetonitrile and 0.1% TFA);
(b) Cyano Column (if product is hydrophobic), 5 micron particle size, 4.6 mm width X 250 mm length, 100 Angstrom pore size, flow=1.0 ml/minute, gradient=95%A/5%B->0%A/100%B in 30 minutes;
(c) Silica Normal Phase Column (if product is hydropholic), 5 micron particle size, 4.6 mm width X 250 mm length, 100 Angstrom pore size, flow=1.0 ml/minute, gradient=0%A/100%B->1000%A/0%B in 30 minutes;

2. Mass Spec for peptides and amino acids can be performed by ESI (electrospray ionization (+ or - mode)) or MALDI (Matrix Assisted Laser Desorption Ionization (+ or - mode)), however, results must be +/- 1.0 Dalton of the calculated theoretical average mass. No impurity displayed in the mass spectrum can exceed 25% abundance.

3. For Mass Spec of GMP peptides LC/MS/MS must be run with one of the above 3 HPLC run protocols and the sequence can be confirmed by MS/MS (Edman degradation is optional, however, but can be expensive).

4. For small organic molecules and solvents can be determined by GC/MS as follows: Column=DB5 (5% Phenylmethylsilicone), 30 meters long, 0.32 mm i.d., ultrapure grade Helium flow=1.0->2.0 ml/minute, 25:1 split, 40 degrees C for 6 minutes, 20 degrees C/minute temperature increase, 250 C degrees for 10 minutes, Detector=280 degrees C.

5. For GMP products, product purity must be >98.0% and raw material purity must be >99.0%, no impurity can be >1.0%, and all impurities >0.1% must be identified by LC/MS/MS or GC/MS.

6. For Capillary Electrophoresis analysis, 0.2 milliMolar Sodium Tetraborate (pH=9.3), Silica Capillary Column=105 cm. length X 50 um I.D., a voltage gradient of 10.0 K.V.->30 K.V. in 20 minutes and hold 30 K.V. for 10 minutes (30 minute run time total), a 5 minute preconditioning step with the Borate buffer, and a detection setting at 220 nm should be used. The sample can be dissolved in 50% Buffer A/50% Buffer B (see NOTE #1 above for the components of "Buffer A & B), Formic Acid, or TFA (Trifluoroacetic Acid).

7. For Antibody work, the peptide purity must be at least 70% by HPLC (as depicted above)and/or Capillary Electrophoresis (optional) to elicit a good immune response from the host animal).

8. For Amino Acid Analysis, the acceptable analytical Peptide Content results will be +/- 5.0% of the theoretically calculated Peptide Content, and the individual amino acid components will be +/- 10% of the theoretically calculated numbers of amino acids in the sequence (unless otherwise partially or full destroyed in the hydrolysis steps of the analysis, upon which, a notation on the certificate of analysis would reflect this condition).

9. For net peptide weight, amino acid analysis must be performed to derive the Peptide Content. The peptide gross weight contains peptide + salts + water (moisture) + residual solvents. Thus gross peptide weight to be measured=Amount of (peptide desired/peptide content)*100.
