Molecular biology

Gene: maps to 12q24.33 (Puente X.S. et al., 1998).
mRNA: size: 2.7-kb. A minor 7.5-kb has been described, probably corresponding to an alternative transcript generated by differential posttranscriptional processing of the MMP-17 mRNA (Puente X.S. et al., 1996). More recently, Kajita M. et al. (1999) described two types of MMP17 transcripts. The minor one corresponded to the cDNA reported by Puente et al. and failed to express protein, while the other was the major transcript with an extended open reading frame and expressed 67 and 71 kDa translation products.
Protein: a 518-aminoacids (aa) polypeptide. The predicted aa sequence displays a similar organization as the remaining MMPs, including a prodomain with the activation locus, the zinc-binding site, and the hemopexin domain. In addition, it contains a C-terminal extension, rich in hydrophobic residues and similar in size to those present in the different membrane-type MMPs (MMP14, MMP15, MMP16) (Puente X.S. et al., 1996).

Breast cancer

Cell lines:
- By RT-PCR analysis, MMP17 expression wwas found in MDA-MB-231, MDA-MB-435, T-47D, ZR-75-1, and Hs578T breast cancer cell lines (Puente X.S. et al., 1996).

- Treatment with collagen type I increassed the steady-state MT1-MMP (MMP14) mRNA levels in invasive BCC but did not induce either MMP14 expression or MMP2 activation in noninvasive BCC lines, which retain epithelial features. Basal MT3-MMP (MMP16) mRNA expression had a pattern similar to that of MMP14 but was not up-regulated by collagen. MMP17 mRNA was seen in both invasive and noninvasive BCC lines and was also not collagen-regulated, and MT2-MMP (MMP15) mRNA was not detected in any of the BCC lines tested (Gilles C. et al., 1997).

Tumors:
- by RT-PCR analysis, MMP17 expression wwas found in 6/6 examined breast carcinomas (Puente X.S. et al., 1996).
