CIA ( Chloroform:Isoamyl alcohol=24:1), Fisher/Sigma
T108.0 ( = 10 mM Tris buffer, pH8.0 )
Procedure
Caution! Use glass or polypropylene tube or pipet with chloroform-containing reagent. Do not use polycarbonate materials which would be destroyed by chloroform!
Dissolve dry pellet in 5ml T108.0.
Extract with 5ml 1:1 phenol/CIA.
Separate phases at 3000 rpm for 10 min at RT (in 15 ml centrifuge tube)
Remove aqueous phase (which contains DNA) to fresh 50 ml centrifuge tube
Add 250ul 10M ammonium acetate to make 0.5M ammonium acetate final concentration, mix
Add 2 volume 200 proof ethanol.
Vortex. Put on dry ice for 20 min (or put in -80oC freezer for >=2 hr w/ 1M ammonium acetate (final) to improve precipitation efficiency).
Spin 15 min at RT at >=15,000 X g ( 24,000 X g works fine)
Decant ethanol
Add 15ml 80% ethanol, vortex
Repellet by centrifugation ( as in step 8 )
Remove ethanol completely
Lyophilize in SpeedVac ( make some holes on parafilm on top of the tube)
