Preparation of Collagen-Coated Plates

Materials

  1. BD collagen solution
  2. PBS
  3. 6 or 12-well plate
  4. Sterile MilliQ H2O
  5. Ice
  6. Pipette and tips
  7. 50 ml sterile Falcon tubes
  8. Laminar flow hood
  9. Tissue culture incubator

Procedure

  1. Cool sterile PBS and Collagen solution on ice.
  2. Under tissue culture hood, in 50 ml tube, dilute collagen with cold PBS to 300ug/ml and keep on ice.
  3. Pour 1.5 ml (for 6-well plate) or 0.65ml (for 12-well plate ) per well.
  4. Shake plate to cover the bottom evenly.
  5. Put plate in 37oC incubator . allow 2hr to overnight to gel.
  6. Place plate in hood and remove cover to let the collagen gels dry down completely overnight (may take up to 24-48hr)
  7. Gently wash off salt precipitates with several (3~5) changes (~30min each) of sterile MilliQ H2O.
  8. At last wash, aspirate H2O as completely as possible and let plate dry completely.

 

Note:

The dried plate can be taped and stored at RT for several weeks.
