To catch a note, you need to press the button that corresponds to the note location (i.e. square or L1 for left, triangle or R1 for middle, and circle or R2 for right) as shown in the picture to the left.  Catch a chain of notes in a bar without missing a beat and you capture that instrument for the rest of the song section, which means you can move on to the next instrument and hear the one you just captured without having to play it.
The game takes place inside a psychdelic octagonal tunnel (for those who have played Tempest back in the '80s, this should be familiar), through which you are travelling.  This is the song that you are playing, with each side of the octagon corresponding to an instrument (either drums, bass, synth, guitar, vocals, scratch, or axe).  See those blue orb things in the drum track to the right?  Those are the notes that you have to play, or "catch', using your catcher; the notes have to be caught right when they are in the catcher.

Successfully capturing bars nets you points and adds to your energy bar (lower left of the screen), which you must keep at least partially filled or you'll die (you'll be booed, but I guess for a DJ that's very bad).  You must make it through the entire song without dying in order to clear it, and you must collect enough points along the way to unlock bonus stages.

That's it.  Those are the basics.  Go rent or buy this game now.

Note:  These screenshots do not match the released game's screens.  For one, the song map on the right of the screen is not lettered in the released version, and the numbers in the lower left are also replaced by power-up icons.
