Suicide gene therapy is based on the
intracellular delivery of a gene coding for an enzyme that transforms
a prodrug into a cytotoxic product, which was suggested in the late
1980s. This approach has two alternatives: toxin gene therapy, in
which the genes for toxic products are transfected directly into tumor
cells; and enzyme-activationg prodrug therapy, in which the transgenes
encode enzymes that activate specific prodrugs to create toxic products.
