To impart the knowledge of domestic animal histology for the veterinary students
The illustrative atlas of human histology is sufficient rather than to conceive valuable background details of veterinary histology.
To understand the normal histo-morphological architexure of different species, in relation with their normal and abnormal conditions.
It is one of the important tools in the hand of practioners and for their diagnostic clues.
METHOD OF STUDY OF HISTOLOGY
A variety of methods were adapted to gain the knowledge of histology. However, some of them dealing with dead cells and some of them dealing with living cells. Some types of cells can be conveniently studied by placing them directly on slides for staining and observed under microscope. For example blood smears. But most histological work, it is very essential to cut or slice the organs or tissues in a few millimeter thickness. And further the tissue is subjected to various technical procedures and histological techniques before the microscopic examination of tissue.z
1. Collection of tissues
2. Preservation of tissues or Fixation of tissue with suitable fixatives
3. Dehydrate the tissues - with suitable chemicals
4. Embedding of tissue with paraffin
5. Sectioning of tissues - employed with different kind of instruments
6. The sections of tissues are affixed to the slides and stained- Staining is done by using different dyes or combination of various colored dyes in order to achieve the contrast between the various components of a tissue between the nucleus and cytoplasm of the cell.
7. Mounting of tissues with DPX. There after the slides are subjected for microscopical examination with different magnifications.
A number of other special methods are used in the study of tissues like
1. Histochemical methods
2. Radio-autography methods
3. Fluorescence microscopy
4. Immunofluorescent techniques
5. Micro-incineration
6. Micro-dissection
7. Immunocytochemistry
8. Enzyme Histochemistry
9. Electronmicroscopy
10. Cyrostat - Frozen sectioning
