Barret: Her adoptive father. He seems very maternal when around her. Marlene seems to be his weakness. In Final Fantasy VII when they narrowly escaped from the Sector 7 Slums where the pillar fell, he got on his knees and screamed, "MARLENE!" He cried hysterically when he discovered she survived.

Aeris: Aeris is the reason Marlene is still alive. If she hadn't taken her to Elmyra's house, she would have been abolished in the destruction of Sector 7.

Tifa: It was revealed that Tifa and Marlene will have an interesting connection in the upcoming sequel. She is posed as a maternal figure to her, as she takes gentle care of all the children that were plagued by Geostigma.

Denzel: Denzel is Marlene's close friend in Final Fantasy VII: Advent Children. She comforts him through his illness and they seem to be about the exact same age.
