Title: PCR-RFLP analysis of Caprine (Goat) Mitochondrial
DNA D-loop polymorphism
Duration:
14 months and previous one year experience as first year trainee
Work:
- Selection of primers from publication and
verification of its alignment with available sequences in the Genbank
using PubMed BLAST search
- Selection of REs based on restriction map
produced with already available sequences with Gene Tool Lite software
- Isolation of genomic DNA from peripheral
blood
- Quantification and standardisation of DNA
template
- PCR amplification of D-loop region of mtDNA
with specific primers
- Verification of amplicon on 1.2% agarose gel
- Digestion of PCR products with Restriction
Enzymes HaeIII, HpaII, HinfI
- Running of the digest on 1.5% agarose gel
- Recording the image and finding out the
different patterns using QUANTITY ONE (Bio-Rad) software
- Finding out the association of different
patterns with economic traits
Loaction: The research was conducted at the Core Laboratory
of Animal Genetics Department of Madras Veterinary College. The lab is one of
the three leading molecular genetics laboratories commissioned by the National
Bureau of Animal Genetic Resources, India for molecular characterization of
animal breeds of India as per the guidance of FAO. Our college is 100
years old and one of the reputed institutions in India.

Biotechnology, Government of India in association with our university
