PC-PLC is essential for TRAIL, Fas and TNF-induced apoptosis

PC-PLC, a membrane lipid, was reported to be an important lipid to mediate signal transduction, but its role in apoptosis is still debated and poorly studied.  PC-PLC as an lipid degrading enzyme when activated can induce formation of many other  lipids, for example DAG, which is one of the production of PC-PLC, is also known reported that can induce many signals in hematopoietic cells, such as PKC.  In our present study, we could see an induction of PC-PLC activity by stimulation of TRAIL, Fas and TNF, which later we found that PC-PLC activity could increase production of DAG.  Apoptosis signal of TRAIL, Fas and TNF can be mediated by DAG to PKCd, which later will induce both extrinsic and intrinsic apoptosis pathways.  Inhibitor of PC-PLC, D609, was applied and our data showed that it could inhibit TRAIL, Fas and TNF-induced apoptosis.  Cardiolipin which is an important mitochondrial membrane lipid was also shown in our study, that D609 could inhibit TRAIL, Fas and TNF-induced cardiolipin oxidation.
