Embedding of 9.5dpc mouse embryos

Harvest 9.5dpc mouse embryos
Fix in 4% paraformaldehyde in PBS for iSmin at 4oC
Wash in rinse buffer for 5min x3
Stain in X-gal staining buffer 2-4hr at 37oC
Post-fix in 4% paraformaldehyde overnight at 4oC
Take photographs if desired
50% ethanol/saline (0.83% NaCl) overnight at 4oC
70% ethanol overnight at 4oC (can be stored for a long time)
90% ethanol l0min r.t.
100% ethanol 15-20min r.t. x2
Toluene l0min x2 (check whether the embryos are cleared)
Transfer the embryos to the molten wax at 60oC for l0min
Pour the wax away
Add new wax, wait for l0min
Pour the wax away
Add new wax, wait for l0min
Embed the embryos directly without changing the wax

Preparation of embryo dish:
Clean with Kiniwipe
Wipe a thin layer of glycerol onto the surface
Place in 60oC oven
